Description

ST3GAL6 encodes a Golgi membrane protein that catalyzes the transfer of sialic acid from CMP-sialic acid to galactose-containing substrates via an alpha-2,3 linkage. This sialyltransferase is involved in the biosynthesis of sialyl-Lewis X and other sialylated glycoconjugates, playing roles in cell-cell adhesion, immune response, and cancer metastasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer Altered sialylation of cell surface glycoproteins promotes tumor invasion and metastasis. PMID: 25691885
Pancreatic Cancer Upregulation of ST3GAL6 enhances sialyl-Lewis X synthesis, facilitating hematogenous metastasis. PMID: 24987072
Acute Myeloid Leukemia ST3GAL6 expression correlates with poor prognosis and may modulate leukemic cell adhesion. PMID: 25982141

Protein Summary

ST3GAL6 is a type II membrane protein localized to the Golgi apparatus. It contains a short N-terminal cytoplasmic domain, a transmembrane region, and a large C-terminal catalytic domain facing the Golgi lumen. The enzyme transfers sialic acid to terminal galactose residues on glycoproteins and glycolipids, forming alpha-2,3 linkages. Its activity is critical for the synthesis of sialyl-Lewis X, a ligand for selectins involved in leukocyte trafficking and cancer cell metastasis.
