Description
SSTR5 encodes a G protein-coupled receptor for somatostatin, a peptide hormone that inhibits the release of growth hormone, insulin, glucagon, and other hormones. SSTR5 is predominantly expressed in the pituitary gland, pancreas, and gastrointestinal tract. It mediates somatostatin's inhibitory effects on hormone secretion and cell proliferation. The receptor is a key therapeutic target for somatostatin analogs used in acromegaly, Cushing disease, and neuroendocrine tumors.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Acromegaly | SSTR5 activation by somatostatin analogs suppresses growth hormone secretion from pituitary adenomas; reduced SSTR5 expression correlates with treatment resistance. | PMID: 25636053, ClinVar |
| Cushing Disease | SSTR5 is the predominant somatostatin receptor subtype in corticotroph adenomas; pasireotide, a multi-receptor somatostatin analog with high SSTR5 affinity, reduces ACTH secretion. | PMID: 22535969, ClinVar |
| Neuroendocrine Tumors (NETs) | SSTR5 overexpression in NETs enables imaging and targeted therapy with radiolabeled somatostatin analogs (e.g., 177Lu-DOTATATE). | PMID: 28289288, COSMIC |
| Pituitary Adenoma | Somatic mutations and altered SSTR5 expression contribute to tumor growth and hormone hypersecretion. | PMID: 30093543, COSMIC |

Protein Summary
SSTR5 is a 364-amino acid G protein-coupled receptor with seven transmembrane domains. It binds somatostatin-14 and somatostatin-28 with high affinity. Upon activation, it couples to Gi/o proteins to inhibit adenylate cyclase, reduce cAMP levels, and modulate ion channels. SSTR5 also activates MAPK and PI3K pathways, influencing cell growth and apoptosis. The receptor undergoes agonist-induced internalization and desensitization. Its expression is regulated by transcription factors including POU1F1 and GATA-2.
