Description
SSTR2 encodes somatostatin receptor 2, a G protein-coupled receptor for somatostatin-14 and -28. It mediates inhibition of hormone secretion, cell proliferation, and induces apoptosis. SSTR2 is highly expressed in neuroendocrine tissues and is a primary target for diagnostic imaging and peptide receptor radionuclide therapy (PRRT) in neuroendocrine tumors.

| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neuroendocrine tumors (NETs) | SSTR2 overexpression enables targeted imaging and therapy via somatostatin analogs | PMID: 31383879 |
| Acromegaly | SSTR2 mediates somatostatin-induced suppression of GH secretion; loss of SSTR2 correlates with resistance to somatostatin analogs | PMID: 25636053 |
| Gastroenteropancreatic NETs | SSTR2 expression is a predictive biomarker for response to octreotide and lanreotide | PMID: 28854169 |

Protein Summary
Somatostatin receptor 2 (SSTR2) is a 369-amino acid transmembrane GPCR that preferentially binds somatostatin-14 and -28. Upon activation, it couples to Gi/o proteins to inhibit adenylyl cyclase, reduce cAMP, and modulate ion channels. SSTR2 is a key mediator of somatostatin's anti-secretory and anti-proliferative effects, and its overexpression in neuroendocrine tumors is exploited clinically for imaging (68Ga-DOTATATE PET) and therapy (177Lu-DOTATATE PRRT).
