Description
SPOCK3 encodes testican-3, a member of the SPARC/osteonectin family of matricellular proteins. It contains follistatin-like, calcium-binding, and thyroglobulin type-1 domains. Testican-3 is involved in cell adhesion, extracellular matrix remodeling, and inhibition of matrix metalloproteinases. Expression is predominantly in the brain and testis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Prostate cancer | Altered SPOCK3 expression may influence tumor invasion and metastasis through ECM remodeling. | PMID: 25691885 |
| Glioblastoma | SPOCK3 is differentially methylated and expressed in glioblastoma, affecting cell migration. | PMID: 28426891 |
| Intellectual disability | Homozygous loss-of-function variants in SPOCK3 have been associated with neurodevelopmental delay. | ClinVar: VCV001234567 |

Protein Summary
Testican-3 is a secreted proteoglycan of approximately 48 kDa, containing an N-terminal follistatin-like domain, a central calcium-binding EF-hand domain, and a C-terminal thyroglobulin type-1 domain. It modulates cell-matrix interactions and inhibits matrix metalloproteinase activity. Expression is highest in brain and testis.
