Description

SOX5 encodes a member of the SOX (SRY-related HMG-box) family of transcription factors, characterized by a conserved high-mobility group (HMG) DNA-binding domain. SOX5 plays critical roles in embryonic development, particularly in chondrogenesis (cartilage formation) and neurogenesis. It acts as a transcriptional activator or repressor depending on context and partner proteins. Mutations in SOX5 are associated with Lamb-Shaffer syndrome (LAMSHF), a neurodevelopmental disorder characterized by intellectual disability, speech delay, and facial dysmorphism. SOX5 is also implicated in several cancers, including glioblastoma and melanoma, where it can promote tumor progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Lamb-Shaffer syndrome (LAMSHF) Haploinsufficiency of SOX5 due to heterozygous loss-of-function mutations or deletions leads to impaired neurodevelopment and craniofacial abnormalities. ClinVar, OMIM #616803
Intellectual disability, autosomal dominant 32 De novo missense or nonsense mutations in SOX5 disrupt DNA binding and transcriptional activity, causing cognitive impairment. ClinVar, OMIM #604975
Glioblastoma SOX5 overexpression promotes glioma cell proliferation and invasion via regulation of target genes involved in cell cycle and migration. COSMIC, PubMed studies
Melanoma SOX5 acts as a lineage-specific oncogene, driving melanoma cell proliferation and metastasis by modulating MITF and other targets. COSMIC, PubMed studies

Mutation functional classification

Loss of Function (LOF)

Nonsense, frameshift, and missense mutations that reduce or abolish SOX5 DNA-binding or transactivation activity; cause Lamb-Shaffer syndrome and intellectual disability.

Gain of Function (GOF)

Gene amplification or overexpression in cancers (glioblastoma, melanoma) leading to enhanced transcriptional activity and oncogenic signaling.

Dominant Negative (DN)

Not well documented; some missense variants may interfere with wild-type SOX5 function in heterodimers, but evidence is limited.

Pathways

• Chondrogenesis and skeletal development (Reactome: R-HSA-5619507)
• SOX-mediated regulation of neural crest cell migration (Reactome: R-HSA-5619508)
• Transcriptional regulation by SOX5 in glioblastoma (KEGG: hsa05214)
• Melanogenesis and MITF regulation (KEGG: hsa04916)

Protein Summary

SOX5 is a 763-amino acid transcription factor containing an N-terminal HMG box DNA-binding domain, a coiled-coil dimerization domain, and a C-terminal transactivation domain. It binds DNA as a monomer or dimer and interacts with partner transcription factors (e.g., SOX6, SOX9) to regulate target genes. SOX5 is essential for chondrogenesis, where it cooperates with SOX6 and SOX9 to activate collagen type II and aggrecan expression. In the nervous system, it controls neuronal migration and differentiation. Post-translational modifications include phosphorylation and sumoylation, which modulate its activity and stability.
