Description

SOCS5 (Suppressor of Cytokine Signaling 5) is a protein-coding gene that encodes a member of the suppressor of cytokine signaling (SOCS) family. SOCS proteins are key negative regulators of cytokine receptor signaling via the JAK-STAT pathway. SOCS5 specifically inhibits signaling by cytokines such as IL-4, IL-6, and EGF by binding to JAK kinases or cytokine receptors, targeting them for proteasomal degradation. It is involved in immune regulation, cell growth, and differentiation. Aberrant SOCS5 expression has been implicated in inflammatory diseases, autoimmune disorders, and certain cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Asthma SOCS5 modulates IL-4 signaling, affecting Th2 cell differentiation and airway inflammation PMID: 15294900
Rheumatoid Arthritis Altered SOCS5 expression in synovial tissue may contribute to chronic inflammation via dysregulated JAK-STAT signaling PMID: 20015550
Hepatocellular Carcinoma SOCS5 downregulation is associated with increased STAT3 activation and tumor progression PMID: 23541953
Prostate Cancer SOCS5 promoter hypermethylation leads to loss of expression and enhanced oncogenic signaling PMID: 21573185

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., p.Leu241fs) are predicted to truncate the SOCS box domain, impairing ubiquitin-mediated degradation of target proteins and leading to sustained cytokine signaling.

Gain of Function (GOF)

No gain-of-function mutations have been reported for SOCS5 in curated databases.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for SOCS5.

Pathways

• JAK-STAT signaling pathway (KEGG: hsa04630)
• Cytokine-cytokine receptor interaction (KEGG: hsa04060)
• EGFR tyrosine kinase inhibitor resistance (KEGG: hsa01521)
• IL-4 signaling pathway (Reactome: R-HSA-6785807)

Protein Summary

SOCS5 is a 536-amino acid protein containing an N-terminal SH2 domain and a C-terminal SOCS box. The SH2 domain binds phosphorylated tyrosine residues on cytokine receptors or JAK kinases, while the SOCS box recruits E3 ubiquitin ligase components to target bound proteins for proteasomal degradation. SOCS5 is predominantly cytoplasmic and is expressed in immune tissues, lung, and liver. It plays a critical role in fine-tuning cytokine responses, particularly in Th2-mediated immunity and growth factor signaling.
