Description
SOCS2 (Suppressor of Cytokine Signaling 2) encodes a member of the SOCS (suppressor of cytokine signaling) family, which are negative regulators of cytokine receptor signaling via the JAK/STAT pathway. SOCS2 inhibits signaling by binding to phosphorylated tyrosine residues on cytokine receptors and targeting associated JAK kinases for proteasomal degradation. It plays a critical role in growth hormone (GH) and insulin-like growth factor 1 (IGF1) signaling, and its dysregulation is linked to growth abnormalities and various cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Growth hormone insensitivity with immune dysregulation | Loss-of-function mutations in SOCS2 lead to enhanced GH signaling, resulting in growth abnormalities and immune defects. | PMID: 15630409 |
| Breast cancer | SOCS2 overexpression is associated with poor prognosis and may promote tumor growth via altered JAK/STAT signaling. | PMID: 17634559 |
| Prostate cancer | SOCS2 hypermethylation and reduced expression correlate with disease progression and metastasis. | PMID: 23361059 |
| Colorectal cancer | SOCS2 downregulation is linked to increased STAT3 activation and tumor progression. | PMID: 22179823 |

Mutation functional classification
Loss of Function (LOF)
Most reported SOCS2 mutations are loss-of-function, impairing its ability to downregulate JAK/STAT signaling, leading to enhanced cytokine responses.
Gain of Function (GOF)
Gain-of-function mutations in SOCS2 are rare and not well characterized; overexpression in some cancers may mimic gain-of-function effects.
Dominant Negative (DN)
No dominant-negative mutations have been reported for SOCS2.

Protein Summary
The SOCS2 protein (UniProt O14508) is 198 amino acids long and contains an N-terminal SH2 domain that binds phosphorylated tyrosine residues on cytokine receptors, and a C-terminal SOCS box that recruits the ubiquitin-transferase machinery. It functions as a key negative feedback regulator of JAK/STAT signaling, particularly downstream of growth hormone and prolactin receptors. SOCS2 is localized in the cytoplasm and nucleus, and its expression is induced by cytokine stimulation.
