Description

SLC39A1 (solute carrier family 39 member 1) encodes the zinc transporter ZIP1, which mediates cellular uptake of zinc from the extracellular space. Zinc is an essential trace element required for numerous enzymatic, structural, and regulatory functions. ZIP1 is ubiquitously expressed and plays a critical role in maintaining intracellular zinc homeostasis. Dysregulation of SLC39A1 has been implicated in prostate cancer, where loss of ZIP1 expression leads to zinc accumulation defects and malignant transformation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate Cancer Loss of ZIP1 expression reduces intracellular zinc, promoting tumorigenesis via altered citrate metabolism and increased oxidative stress. PMID: 14612522; NCBI GeneRIF
Zinc Deficiency Impaired ZIP1 function may contribute to systemic zinc deficiency, affecting immune function and growth. OMIM #604731; UniProt

Protein Summary

ZIP1 is a transmembrane protein with eight predicted transmembrane domains, localized to the plasma membrane and intracellular vesicles. It functions as a homodimer to import zinc into the cytosol. The protein contains a histidine-rich motif essential for zinc binding and transport. ZIP1 is regulated at transcriptional and post-translational levels by zinc availability.
