Description
SKIC2 encodes a component of the SKI complex, which is essential for RNA exosome-mediated degradation of aberrant RNAs. The protein is a putative RNA helicase that unwinds RNA structures to facilitate exosome access. Mutations in SKIC2 are associated with trichohepatoenteric syndrome, a rare autosomal recessive disorder characterized by intractable diarrhea, liver disease, and immune abnormalities.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Trichohepatoenteric syndrome | Loss-of-function mutations impair RNA surveillance, leading to accumulation of defective RNAs and cellular stress | OMIM #222470; PMID: 22521418 |
| Syndromic diarrhea | Defective SKI complex disrupts RNA exosome function, causing intestinal epithelial dysfunction | OMIM #614602 |

Protein Summary
SKIC2 (also known as SKIV2L2 or MTR4) is a 1,022-amino acid protein containing a DExH-box helicase domain. It is a core subunit of the SKI complex, which recruits the RNA exosome to degrade aberrant RNAs. The protein localizes to both the nucleus and cytoplasm and is essential for RNA quality control.
