Description
SIGLEC11 (sialic acid binding Ig-like lectin 11) is a member of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It is a transmembrane receptor that recognizes sialic acid-containing glycans. SIGLEC11 is predominantly expressed in human microglia and plays a role in neuroimmune regulation, including inhibition of inflammatory responses. It has been implicated in several neurological and psychiatric conditions, as well as in cancer biology.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Schizophrenia | Altered SIGLEC11 expression in microglia may affect synaptic pruning and neuroinflammation, contributing to schizophrenia pathophysiology. | Case-control studies show differential expression in brain tissue (e.g., PMID: 25816173). |
| Alzheimer's disease | SIGLEC11-mediated inhibition of microglial phagocytosis may impair clearance of amyloid-beta plaques. | Expression changes observed in AD brains; functional studies in vitro (e.g., PMID: 27466428). |
| Cancer (various) | SIGLEC11 expression on tumor-associated macrophages may modulate tumor immune evasion via sialic acid interactions. | Immunohistochemistry and transcriptomic data from COSMIC and literature (e.g., PMID: 29217573). |

Mutation functional classification
Loss of Function (LOF)
Loss-of-function mutations in SIGLEC11 have not been extensively reported; however, reduced expression or function may impair anti-inflammatory signaling in microglia.
Gain of Function (GOF)
Gain-of-function mutations are not well documented; overexpression in certain cancers may enhance immune evasion.
Dominant Negative (DN)
No dominant-negative mutations have been described for SIGLEC11.

Protein Summary
SIGLEC11 is a type I transmembrane protein with an extracellular N-terminal V-set domain that binds sialic acid, followed by two C2-set domains. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in its cytoplasmic tail, which recruit phosphatases to dampen immune activation. In the brain, SIGLEC11 is selectively expressed in microglia and contributes to maintaining an anti-inflammatory environment. It also has roles in cell-cell interactions and may be involved in tumor immune evasion.
