Description
SEMA3F (Semaphorin 3F) is a protein-coding gene that belongs to the class 3 semaphorin family. It encodes a secreted glycoprotein that functions as a chemorepellent for axons and as an inhibitor of angiogenesis and tumor growth. SEMA3F signals through neuropilin-1 and neuropilin-2 receptors and plays a critical role in neural development, immune regulation, and cancer suppression. Loss of SEMA3F expression is frequently observed in various cancers, contributing to tumor progression and metastasis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Lung Cancer | Loss of SEMA3F expression due to promoter methylation or deletion at 3p21.31 leads to increased tumor cell migration and invasion. | PMID: 15696293, COSMIC |
| Breast Cancer | Reduced SEMA3F expression correlates with poor prognosis and promotes angiogenesis via VEGF pathway. | PMID: 20010872, ClinVar |
| Colorectal Cancer | SEMA3F downregulation enhances epithelial-mesenchymal transition (EMT) and metastasis. | PMID: 23934199 |
| Neuroblastoma | SEMA3F acts as a tumor suppressor; its loss promotes neuroblastoma cell proliferation. | PMID: 18794855 |
| Axon Guidance Defects | SEMA3F mutations disrupt repulsive signaling, leading to abnormal neuronal pathfinding. | OMIM: 601124 |

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations that truncate the protein, as well as promoter hypermethylation leading to silencing, result in loss of SEMA3F function.
Gain of Function (GOF)
No gain-of-function mutations have been reported for SEMA3F.
Dominant Negative (DN)
No dominant-negative mutations have been characterized for SEMA3F.

Protein Summary
SEMA3F encodes a 785-amino-acid secreted semaphorin with a conserved Sema domain, a PSI domain, and an immunoglobulin-like domain. The protein forms homodimers and binds to neuropilin-2 (NRP2) with high affinity, and to neuropilin-1 (NRP1) with lower affinity. SEMA3F signaling through plexin co-receptors induces cytoskeletal collapse and growth cone repulsion in neurons. In non-neuronal tissues, SEMA3F inhibits integrin-mediated adhesion and suppresses tumor angiogenesis by competing with VEGF for neuropilin binding. The protein is post-translationally modified by glycosylation and proteolytic cleavage.
