Description
SEC22B encodes a member of the SEC22 family of vesicle trafficking proteins. It is a R-SNARE involved in endoplasmic reticulum (ER)-Golgi transport, autophagy, and phagocytosis. SEC22B is also implicated in cross-presentation of antigens in dendritic cells and plays a role in immune evasion by certain viruses.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (various types) | SEC22B overexpression may promote tumor growth by enhancing vesicle trafficking and immune evasion. | COSMIC; PMID: 31073040 |
| Viral infections (e.g., influenza, SARS-CoV-2) | SEC22B is hijacked by viruses to facilitate viral entry and replication. | PMID: 32818467 |
| Neurodegenerative disorders | Dysregulation of SEC22B-mediated ER-Golgi transport may contribute to protein aggregation. | PMID: 29518349 |

Mutation functional classification
Loss of Function (LOF)
Rare missense variants may impair SNARE complex formation and vesicle fusion.
Gain of Function (GOF)
Not well characterized; overexpression in cancer may represent a gain-of-function phenotype.

Protein Summary
SEC22B is a 250-amino acid R-SNARE protein localized to the ER-Golgi intermediate compartment. It forms SNARE complexes with syntaxin-5, GOSR1, and BET1L to mediate vesicle fusion. SEC22B also regulates autophagy by interacting with ATG9A and is critical for cross-presentation of exogenous antigens in dendritic cells.
