Description
SDCBP (syndecan binding protein), also known as syntenin or MDA-9, encodes a scaffold protein that binds to the cytoplasmic domain of syndecans. It plays a key role in exosome biogenesis, cell adhesion, cytoskeletal organization, and intracellular trafficking. SDCBP is overexpressed in multiple cancers and is implicated in tumor progression and metastasis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Melanoma | SDCBP overexpression promotes invasion and metastasis via NF-κB and FAK signaling | PMID: 23934199 |
| Breast cancer | SDCBP enhances exosome secretion and tumor cell migration | PMID: 30523278 |
| Glioma | SDCBP supports cell proliferation and invasion through PDZ domain interactions | PMID: 25605248 |
| Hepatocellular carcinoma | SDCBP upregulation correlates with poor prognosis and metastasis | PMID: 27840973 |

Protein Summary
SDCBP encodes syntenin, a 298-amino acid PDZ domain-containing scaffold protein. It localizes to the plasma membrane and endosomal compartments, where it links syndecans to cytoskeletal and signaling molecules. Syntenin is critical for exosome production and is frequently upregulated in cancers, promoting invasion and metastasis.
