Description
SDC4 encodes syndecan 4, a transmembrane heparan sulfate proteoglycan that functions as a coreceptor for growth factors, cytokines, and extracellular matrix components. It plays critical roles in cell adhesion, migration, cytoskeletal organization, and signal transduction via interactions with integrins and growth factor receptors. SDC4 is implicated in wound healing, angiogenesis, and tumor progression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast cancer | SDC4 overexpression promotes tumor cell invasion and metastasis through activation of FAK and ERK signaling pathways. | COSMIC, ClinVar |
| Colorectal cancer | Altered SDC4 expression correlates with poor prognosis; loss of SDC4 enhances Wnt/β-catenin signaling. | COSMIC, NCBI |
| Osteoarthritis | SDC4 mediates chondrocyte mechanotransduction; dysregulation contributes to cartilage degradation. | OMIM, NCBI |
| Cardiovascular disease | SDC4 modulates endothelial cell function and angiogenesis; variants associated with atherosclerosis risk. | ClinVar, NCBI |

Mutation functional classification
Loss of Function (LOF)
Mutations that reduce cell surface expression or disrupt heparan sulfate chain attachment, impairing coreceptor activity.
Gain of Function (GOF)
Overexpression or mutations that enhance shedding or constitutive activation of downstream signaling (e.g., FAK, ERK).
Dominant Negative (DN)
Truncated or dimerization-defective mutants that interfere with wild-type syndecan 4 function.

Protein Summary
Syndecan 4 is a type I transmembrane protein with an extracellular domain bearing heparan sulfate chains that bind ligands such as FGF, VEGF, and fibronectin. Its cytoplasmic domain interacts with PKCα, syntenin, and cytoskeletal proteins to regulate focal adhesion dynamics and cell migration. The protein is shed from the cell surface by matrix metalloproteinases, generating soluble ectodomains that modulate signaling.
