Description

SDC1 encodes syndecan 1, a transmembrane heparan sulfate proteoglycan that functions as a co-receptor for growth factors, cytokines, and extracellular matrix components. It is involved in cell proliferation, adhesion, and migration. SDC1 is a well-established marker for plasma cells and is frequently dysregulated in multiple myeloma and other cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Multiple Myeloma Overexpression of SDC1 (CD138) on malignant plasma cells promotes cell survival and adhesion in the bone marrow microenvironment. Shedding of the ectodomain correlates with poor prognosis. ClinVar, COSMIC, NCBI
Breast Cancer Altered SDC1 expression influences tumor growth and metastasis through modulation of growth factor signaling (e.g., FGF, VEGF). NCBI, COSMIC
Lung Cancer SDC1 overexpression is associated with increased invasiveness and poor survival in non-small cell lung cancer. COSMIC, NCBI
Hepatocellular Carcinoma SDC1 expression changes affect cell adhesion and Wnt signaling, contributing to tumor progression. NCBI, COSMIC

Loss of Function (LOF)

Loss-of-function mutations in SDC1 are rare and not well characterized; they may impair cell adhesion and signaling.

Gain of Function (GOF)

Gain-of-function is typically associated with overexpression rather than specific activating mutations.

Dominant Negative (DN)

No dominant-negative mutations have been reported for SDC1.

Protein Summary

Syndecan 1 (CD138) is a type I transmembrane heparan sulfate proteoglycan. The core protein (310 amino acids) has an extracellular domain with attachment sites for heparan sulfate and chondroitin sulfate chains, a single transmembrane domain, and a short cytoplasmic tail. It mediates cell-cell and cell-matrix adhesion, acts as a co-receptor for growth factors (e.g., FGF, VEGF), and modulates signaling pathways. Shedding of the ectodomain by matrix metalloproteinases releases soluble syndecan 1, which can regulate tumor microenvironment.
