Description

SCARA3 (scavenger receptor class A member 3) encodes a macrophage scavenger receptor-like protein that functions in the clearance of modified low-density lipoproteins and cellular debris. It is induced by oxidative stress and may act as a tumor suppressor by protecting cells from oxidative damage. The protein is a member of the scavenger receptor family and is involved in host defense and atherosclerosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Atherosclerosis SCARA3 mediates uptake of oxidized LDL by macrophages, contributing to foam cell formation and plaque development. PMID: 10988299
Prostate Cancer SCARA3 expression is downregulated in prostate cancer; loss may promote tumor progression via impaired oxidative stress response. PMID: 15126332
Breast Cancer Reduced SCARA3 expression correlates with poor prognosis; potential tumor suppressor role through ROS clearance. PMID: 19137010

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations in SCARA3 are predicted to cause loss of scavenger receptor function, impairing clearance of oxidized LDL and oxidative stress response.

Gain of Function (GOF)

No gain-of-function mutations have been reported for SCARA3.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for SCARA3.

Protein Summary

SCARA3 is a 372-amino acid type II transmembrane glycoprotein with a collagen-like domain and a scavenger receptor cysteine-rich (SRCR) domain. It forms trimers and binds a broad range of polyanionic ligands, including oxidized LDL, bacteria, and apoptotic cells. The protein is localized to the plasma membrane and endosomes, and its expression is upregulated by oxidative stress, suggesting a protective role against cellular damage.
