Description

S100A7A (S100 calcium binding protein A7A) is a member of the S100 family of EF-hand calcium-binding proteins. It is located in the epidermal differentiation complex on chromosome 1q21.3. S100A7A is involved in innate immune responses, inflammation, and keratinocyte differentiation. It is upregulated in psoriatic skin and certain cancers, including breast and lung cancer, where it may promote tumor progression and metastasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Psoriasis Overexpression in psoriatic epidermis; promotes inflammation via chemotaxis and cytokine induction PubMed, NCBI Gene
Breast cancer Upregulated in invasive breast carcinoma; associated with poor prognosis PubMed, COSMIC
Lung cancer Elevated expression in non-small cell lung cancer; linked to metastasis PubMed, COSMIC

Protein Summary

S100A7A is a 101-amino acid protein (11.4 kDa) containing two EF-hand calcium-binding domains. It functions as a homodimer and can also heterodimerize with S100A7. The protein is secreted and acts as a chemoattractant for immune cells, particularly neutrophils and T cells. It binds to the receptor for advanced glycation end products (RAGE) and Toll-like receptor 4 (TLR4), activating NF-κB and MAPK signaling pathways. S100A7A is implicated in antimicrobial defense, wound healing, and tumorigenesis.
