Description

RPLP2 encodes the ribosomal protein P2, a component of the 60S large ribosomal subunit. The protein forms part of the lateral stalk structure that binds elongation factors and is critical for efficient mRNA translation. RPLP2 is highly conserved and expressed in all eukaryotic cells. Its expression is dysregulated in several cancers and it is a known autoantigen in systemic lupus erythematosus.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Systemic lupus erythematosus Autoantibodies against ribosomal P proteins (including RPLP2) target the ribosome, contributing to immune complex formation and tissue damage. OMIM 180470; ClinVar
Colorectal cancer Overexpression of RPLP2 promotes protein synthesis and cell proliferation. COSMIC; PubMed studies
Hepatocellular carcinoma Upregulation of RPLP2 correlates with poor prognosis and increased metastatic potential. COSMIC; PubMed studies
Breast cancer RPLP2 is overexpressed in tumor tissues and associated with aggressive subtypes. COSMIC; PubMed studies

Protein Summary

RPLP2 is a 115-amino-acid acidic ribosomal protein (11.7 kDa) that forms a homodimer with RPLP1 and associates with RPLP0 to create the ribosomal stalk. This stalk is essential for recruiting elongation factors eEF1A and eEF2 during translation. The protein contains a conserved C-terminal domain that is highly acidic and mediates interactions with the ribosome. Post-translational modifications include phosphorylation at Ser105, which regulates ribosome binding. RPLP2 is also a major autoantigen in systemic lupus erythematosus, where anti-P antibodies target the C-terminal region.
