Description

RNF4 encodes a member of the RING finger protein family that functions as a SUMO-targeted ubiquitin ligase (STUbL). It specifically recognizes SUMOylated proteins and promotes their ubiquitination and subsequent proteasomal degradation. RNF4 plays critical roles in DNA damage response, homologous recombination repair, and maintenance of genome stability. It is also involved in transcriptional regulation and spermatogenesis.

Mutation functional classification

Loss of Function (LOF)

Missense mutations in the RING domain (e.g., p.Arg114Gln) abolish E3 ubiquitin ligase activity, impairing SUMO-targeted degradation.

Gain of Function (GOF)

Not well characterized; overexpression in prostate cancer may confer gain-of-function by enhancing AR signaling.

Dominant Negative (DN)

Truncating mutations or those disrupting SUMO interaction may act as dominant negatives by sequestering SUMOylated substrates.

Pathways

SUMOylation and ubiquitination crosstalk
Homologous recombination repair
p53 signaling pathway
Androgen receptor signaling

Protein Summary

RNF4 is a 190-amino acid protein containing an N-terminal SUMO-interacting motif (SIM) and a C-terminal RING finger domain. It acts as a SUMO-targeted ubiquitin ligase (STUbL), recognizing SUMOylated proteins and catalyzing their ubiquitination. This activity is essential for the resolution of DNA repair foci, transcriptional regulation, and proteostasis. RNF4 is highly expressed in testis and bone marrow, and its dysregulation is implicated in several cancers.
