Description

RNASEK (ribonuclease K) encodes a conserved endoribonuclease that cleaves single-stranded RNA. The protein is localized to the endoplasmic reticulum and is involved in RNA metabolism, including processing of ribosomal RNA and degradation of specific transcripts. RNASEK has been implicated in cellular stress responses and viral replication, particularly for influenza A virus. Expression is widespread but highest in tissues with high secretory activity.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Influenza A infection RNASEK is required for influenza virus replication; it facilitates viral RNA entry into the cytosol by promoting endosomal escape. PMID: 25533951
Colorectal cancer Overexpression of RNASEK is associated with poor prognosis; may promote tumor growth via RNA processing alterations. PMID: 30872518
Hepatocellular carcinoma Upregulated in liver cancer; potential role in cell proliferation and migration. PMID: 32015556

Protein Summary

Ribonuclease K is a 227-amino acid protein with a single endoribonuclease domain. It localizes to the endoplasmic reticulum and cleaves single-stranded RNA with preference for AU-rich sequences. The protein is highly conserved across vertebrates. RNASEK is essential for influenza A virus infection by mediating endosomal escape of viral ribonucleoproteins. In cancer, overexpression correlates with poor prognosis in colorectal and hepatocellular carcinomas, suggesting a role in tumor progression.
