Description
RGS13 (Regulator of G Protein Signaling 13) is a member of the regulator of G protein signaling (RGS) family. It acts as a GTPase-activating protein (GAP) for Gα subunits of heterotrimeric G proteins, accelerating the termination of G protein-coupled receptor (GPCR) signaling. RGS13 is predominantly expressed in lymphoid tissues, particularly in germinal center B cells, and plays a role in immune cell activation, migration, and differentiation. It has been implicated in allergic inflammation, autoimmune diseases, and certain cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Allergic rhinitis | RGS13 modulates mast cell and basophil degranulation via GPCR signaling; altered expression linked to increased histamine release. | NCBI Gene, OMIM |
| Asthma | RGS13 expression is reduced in airway epithelial cells, leading to enhanced G protein signaling and bronchoconstriction. | NCBI Gene, OMIM |
| B-cell lymphoma | RGS13 is overexpressed in germinal center B-cell lymphomas; may promote cell survival and proliferation through dysregulated GPCR signaling. | COSMIC, NCBI Gene |
| Breast cancer | RGS13 expression is altered in breast cancer tissues; potential role in tumor progression via modulation of chemokine signaling. | COSMIC, NCBI Gene |

Mutation functional classification
Loss of Function (LOF)
Mutations that impair GAP activity (e.g., p.Arg53Cys) lead to prolonged GPCR signaling, potentially contributing to immune dysregulation.
Gain of Function (GOF)
Not well characterized; no clear gain-of-function mutations reported.
Dominant Negative (DN)
Not reported for RGS13.

Protein Summary
RGS13 is a 159-amino acid protein containing a conserved RGS domain that mediates its GAP activity. It accelerates the hydrolysis of GTP bound to Gαi and Gαq subunits, thereby terminating GPCR signaling. RGS13 is highly expressed in germinal center B cells and modulates chemokine receptor signaling, affecting B cell migration and antibody production. Its dysregulation is associated with allergic diseases and lymphomas.
