Description

The RECK gene encodes a membrane-anchored glycoprotein that functions as a negative regulator of matrix metalloproteinases (MMPs), particularly MMP-2, MMP-9, and MT1-MMP. It is widely expressed in normal tissues but frequently downregulated in various cancers, contributing to tumor invasion and metastasis. RECK is considered a tumor suppressor gene.

Protein Summary

The RECK protein is a 971-amino acid glycoprotein anchored to the plasma membrane via a C-terminal glycosylphosphatidylinositol (GPI) anchor. It contains multiple Kazal-like domains that inhibit matrix metalloproteinases (MMPs), including MMP-2, MMP-9, and MT1-MMP. By regulating MMP activity, RECK controls extracellular matrix remodeling, cell migration, and angiogenesis. Loss of RECK expression is a hallmark of many aggressive cancers.
