Description

RCHY1 encodes an E3 ubiquitin-protein ligase that targets p53 for proteasomal degradation, thereby negatively regulating p53-mediated apoptosis and cell cycle arrest. It contains a RING finger domain and a CHY zinc finger domain, and is involved in cellular stress responses, DNA damage repair, and tumorigenesis.

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations that truncate the protein, impairing ubiquitin ligase activity.

Gain of Function (GOF)

Not well documented; overexpression in tumors may act as a gain-of-function by enhancing p53 degradation.

Dominant Negative (DN)

Not reported.

Gene Ontology (GO)

• ubiquitin-protein transferase activity • zinc ion binding
• protein ubiquitination • negative regulation of apoptotic process
• regulation of cell cycle

Pathways

• p53 signaling pathway
• Ubiquitin mediated proteolysis

Protein Summary

RCHY1 (PIRH2) is a 261-amino acid E3 ubiquitin ligase that catalyzes the ubiquitination and subsequent proteasomal degradation of p53. It also interacts with other substrates such as androgen receptor and p73, modulating cell proliferation and survival. The protein is overexpressed in several cancers, contributing to tumor progression and therapy resistance.
