Description

RBPMS (RNA Binding Protein, mRNA Processing Factor) encodes a member of the RNA recognition motif (RRM) family of RNA-binding proteins. It is involved in post-transcriptional regulation, including alternative splicing, mRNA stability, and translation. RBPMS is highly expressed in smooth muscle tissues and plays a critical role in maintaining the contractile phenotype of smooth muscle cells. It also functions as a cofactor for the transcription factor SRF (serum response factor) and is implicated in cancer, particularly in colorectal and breast cancer, where it may act as a tumor suppressor or oncogene depending on context.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer RBPMS downregulation correlates with poor prognosis; may regulate splicing of cancer-related genes PMID: 25605248
Breast Cancer RBPMS expression is reduced in invasive tumors; loss associated with metastasis PMID: 23555266
Smooth Muscle Dysfunction RBPMS knockout in mice leads to impaired smooth muscle contractility PMID: 20086094
Prostate Cancer RBPMS expression is altered in prostate cancer cell lines PMID: 29187737

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations are predicted to cause loss of RNA-binding activity and downstream splicing regulation.

Gain of Function (GOF)

No gain-of-function mutations have been reported in RBPMS.

Dominant Negative (DN)

No dominant-negative mutations have been characterized.

Pathways

• SRF-dependent gene expression (Reactome: R-HSA-525793)
• mRNA Splicing - Major Pathway (Reactome: R-HSA-72163)
• Processing of Capped Intron-Containing Pre-mRNA (Reactome: R-HSA-72203)

Protein Summary

The RBPMS protein (UniProt Q93062) contains a single N-terminal RNA recognition motif (RRM) that mediates binding to RNA targets, particularly in the 3' UTR of mRNAs. It shuttles between the nucleus and cytoplasm and interacts with SRF to regulate smooth muscle-specific gene expression. RBPMS also modulates alternative splicing of transcripts involved in cell proliferation and migration. Its expression is frequently lost in aggressive cancers, suggesting a tumor-suppressive role.
