Description
RBM3 (RNA Binding Motif Protein 3) is a member of the glycine-rich RNA-binding protein family. It is induced by cold shock and plays a role in mRNA stabilization, translational regulation, and alternative splicing. RBM3 is involved in cellular stress responses, neuroprotection, and has been implicated in various cancers, where it can promote cell proliferation and survival.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast cancer | RBM3 overexpression correlates with poor prognosis; promotes proliferation via mRNA stabilization of oncogenes | PMID: 23555253 |
| Colorectal cancer | RBM3 upregulation associated with tumor progression and chemoresistance | PMID: 25944712 |
| Neurodegenerative diseases | RBM3 expression is neuroprotective in models of Alzheimer's and prion disease; enhances synaptic protein synthesis | PMID: 26005863 |
| Ovarian cancer | High RBM3 expression linked to improved survival in serous ovarian carcinoma | PMID: 21947006 |

Mutation functional classification
Loss of Function (LOF)
Frameshift insertions (e.g., c.523_524insA) are predicted to cause loss of RNA-binding activity.
Gain of Function (GOF)
Not well characterized; overexpression in tumors suggests possible gain-of-function in oncogenic contexts.
Dominant Negative (DN)
No dominant-negative mutations reported.

Protein Summary
RBM3 is a 157-amino acid protein containing an N-terminal RNA recognition motif (RRM) and a C-terminal glycine-rich domain. It shuttles between nucleus and cytoplasm, binding to AU-rich elements in 3' UTRs of target mRNAs to enhance their stability and translation. Under cold stress, RBM3 is upregulated and promotes global protein synthesis, contributing to neuroprotection. In cancer, RBM3 overexpression drives tumor growth by stabilizing pro-survival transcripts.
