Description
RASAL2 (RAS protein activator like 2) encodes a member of the RAS GTPase-activating protein (GAP) family. The protein contains a PH domain, a C2 domain, and a GAP domain, and functions as a negative regulator of RAS signaling by stimulating the intrinsic GTPase activity of RAS proteins, converting them from the active GTP-bound state to the inactive GDP-bound state. RASAL2 has been implicated in both tumor suppression and oncogenesis depending on cellular context and isoform expression. It is also involved in cell adhesion, migration, and cytoskeletal organization.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast Cancer | Loss of RASAL2 expression promotes RAS activation and metastasis; alternatively, specific isoforms may act as oncogenes. | PMID: 24909774, 25944712 |
| Lung Cancer | Reduced RASAL2 expression correlates with poor prognosis and increased RAS signaling. | PMID: 27864367 |
| Colorectal Cancer | RASAL2 downregulation is associated with tumor progression and RAS pathway activation. | PMID: 29367600 |
| Ovarian Cancer | RASAL2 expression loss linked to aggressive disease and RAS-MAPK pathway activation. | PMID: 27478017 |

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations (e.g., p.Arg339*, p.Thr734Asnfs*5) lead to truncated protein lacking functional GAP domain, resulting in loss of RAS-GAP activity and increased RAS signaling.
Gain of Function (GOF)
No well-characterized gain-of-function mutations reported in RASAL2; however, overexpression of certain isoforms may promote oncogenic signaling in specific contexts.
Dominant Negative (DN)
No dominant-negative mutations have been described for RASAL2.

Protein Summary
RASAL2 is a 1159-amino acid protein containing an N-terminal PH domain, a C2 domain, and a C-terminal GAP domain. It localizes to the cytoplasm and plasma membrane, where it negatively regulates RAS activity by accelerating GTP hydrolysis. Alternative splicing produces multiple isoforms with potentially distinct functions. RASAL2 is widely expressed, with highest levels in brain and colon. Its dysregulation contributes to cancer progression through both loss- and gain-of-function mechanisms.
