Description

RAMP2 (Receptor Activity Modifying Protein 2) encodes a member of the RAMP family of single-pass transmembrane proteins. RAMP2 forms a functional receptor complex with calcitonin receptor-like receptor (CALCRL) to create the adrenomedullin (AM) receptor. This complex is essential for vascular development, angiogenesis, and regulation of vascular tone. RAMP2 is also involved in lymphangiogenesis and cardiovascular homeostasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Lymphatic malformation RAMP2 loss-of-function impairs adrenomedullin signaling, disrupting lymphatic endothelial cell development PMID: 31133753
Cardiovascular disease RAMP2 deficiency leads to vascular fragility, hypotension, and impaired angiogenesis PMID: 23395172
Preeclampsia Altered RAMP2 expression affects placental adrenomedullin signaling, contributing to hypertension PMID: 21788487

Mutation functional classification

Loss of Function (LOF)

RAMP2 loss-of-function mutations impair adrenomedullin receptor formation, leading to lymphatic and vascular defects.

Gain of Function (GOF)

No gain-of-function mutations reported in RAMP2.

Dominant Negative (DN)

No dominant-negative mutations reported in RAMP2.

Pathways

• Adrenomedullin signaling pathway (Reactome: R-HSA-420092)
• GPCR ligand binding (Reactome: R-HSA-500792)
• Vascular smooth muscle contraction (KEGG: hsa04270)

Protein Summary

RAMP2 is a 175-amino acid single-pass transmembrane protein with an N-terminal extracellular domain and a short cytoplasmic tail. It functions as a chaperone and co-receptor for CALCRL, enabling adrenomedullin binding and signaling. RAMP2 is critical for endothelial cell survival, angiogenesis, and vascular integrity. Its expression is enriched in lung, heart, and placenta.
