Description

RAB31 (RAB31, member RAS oncogene family) is a protein-coding gene that encodes a small GTPase belonging to the RAS superfamily. RAB31 regulates intracellular membrane trafficking, including endocytosis and exocytosis, and is implicated in cell proliferation, migration, and invasion. Overexpression of RAB31 has been observed in several cancers, including breast, lung, and gastric cancers, where it promotes tumor progression and poor prognosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer RAB31 overexpression enhances EGFR recycling and PI3K/AKT signaling, promoting cell proliferation and invasion. PMID: 25609832
Gastric cancer RAB31 upregulation correlates with lymph node metastasis and poor survival; promotes EMT via TGF-β pathway. PMID: 29367643
Lung adenocarcinoma RAB31 is a direct target of miR-215 and its silencing reduces tumor growth and metastasis. PMID: 31073084
Glioblastoma RAB31 is overexpressed and associated with poor prognosis; regulates EGFR trafficking and signaling. PMID: 27562872

Protein Summary

RAB31 is a small GTPase of the Rab family that cycles between an active GTP-bound and inactive GDP-bound state. It localizes to endosomes and the perinuclear region, regulating endocytic trafficking and recycling of receptors such as EGFR. RAB31 promotes cell migration, invasion, and proliferation through activation of PI3K/AKT and MAPK pathways. Its overexpression in multiple cancers correlates with poor prognosis, making it a potential therapeutic target.
