Description

RAB2A is a member of the RAS oncogene family of small GTPases that regulate vesicle trafficking, specifically the transport between the endoplasmic reticulum (ER) and the Golgi apparatus. It cycles between an active GTP-bound form and an inactive GDP-bound form, controlling membrane docking and fusion. RAB2A is also implicated in autophagy, cell proliferation, and tumorigenesis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Altered RAB2A expression and mutations may disrupt vesicular transport and autophagy, promoting tumor growth and metastasis. COSMIC database reports somatic mutations in multiple cancer types.
Neurodegenerative disorders Dysregulation of RAB2A-mediated trafficking may contribute to protein aggregation and neuronal dysfunction. Limited evidence; studies suggest links to autophagy defects.

Protein Summary

RAB2A is a 212-amino-acid small GTPase (23.6 kDa) that localizes to the Golgi apparatus and vesicular structures. It contains conserved GTP-binding domains and a C-terminal prenylation motif for membrane attachment. The protein cycles between active GTP-bound and inactive GDP-bound states, regulated by guanine nucleotide exchange factors (GEFs) and GTPase-activating proteins (GAPs). RAB2A is essential for retrograde transport from the Golgi to the ER and plays a role in autophagosome formation.
