Description
RAB26 is a member of the RAS oncogene family of small GTPases. It functions as a key regulator of vesicle trafficking, particularly in exocytosis and autophagy. RAB26 is involved in the secretion of insulin from pancreatic beta cells and in the regulation of autophagic flux. It is also implicated in cancer progression and neurodegenerative diseases.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Diabetes mellitus (type 2) | RAB26 regulates insulin granule exocytosis in pancreatic beta cells; impaired function leads to reduced insulin secretion. | PMID: 23415227 |
| Breast cancer | RAB26 overexpression promotes tumor growth and metastasis via enhanced exocytosis of pro-invasive factors. | PMID: 28753428 |
| Lung adenocarcinoma | RAB26 mutations and altered expression correlate with poor prognosis and increased metastatic potential. | COSMIC analysis |
| Neurodegenerative disorders (Alzheimer's disease) | RAB26 modulates autophagic clearance of amyloid-beta; dysregulation contributes to protein aggregation. | PMID: 31073057 |

|---|---|---|---|
| c.437G>A (p.Arg146Gln) | Missense | 0.02% (gnomAD) | Alters GTP-binding affinity; potential loss of function |
| c.584T>C (p.Leu195Pro) | Missense | 0.01% (gnomAD) | Disrupts membrane localization; reduced exocytosis |
| c.123_124insA (p.Glu42Argfs*5) | Frameshift | <0.01% (COSMIC) | Truncated protein; loss of function in cancer |
| c.742G>T (p.Glu248*) | Nonsense | <0.01% (COSMIC) | Premature stop; loss of function |
Mutation functional classification
Loss of Function (LOF)
Missense mutations (e.g., p.Arg146Gln) and truncating variants (p.Glu42Argfs*5, p.Glu248*) impair GTP binding or protein stability, reducing vesicle trafficking and autophagy.
Gain of Function (GOF)
Not well-documented; overexpression in breast cancer may act as a gain-of-function by enhancing exocytosis of oncogenic factors.
Dominant Negative (DN)
No confirmed dominant-negative mutations reported; some missense variants may exert dominant-negative effects by sequestering effectors.

Pathways
• RAB26 regulates insulin secretion (Reactome: R-HSA-422085)
• Autophagy - RAB26 modulates autophagosome-lysosome fusion (KEGG: hsa04140)
• Vesicle-mediated transport (Reactome: R-HSA-5653656)
Protein Summary
RAB26 is a small GTPase of the Rab family, localized to secretory vesicles and autophagic membranes. It cycles between an active GTP-bound state and an inactive GDP-bound state, controlling vesicle docking and fusion. In pancreatic beta cells, RAB26 facilitates insulin granule exocytosis. In cancer cells, it promotes secretion of matrix metalloproteinases and growth factors, enhancing invasion. RAB26 also plays a role in autophagy by recruiting tethering factors to autophagosomes. Its dysregulation is linked to diabetes, cancer, and neurodegeneration.
