Description

QRFP (pyroglutamylated RFamide peptide) is a gene that encodes a neuropeptide precursor. The mature peptides (QRFP-43 and QRFP-26) are ligands for the G protein-coupled receptor GPR103 (QRFPR). QRFP is involved in the regulation of appetite, energy balance, and feeding behavior. It is primarily expressed in the central nervous system and peripheral tissues such as the pancreas and adrenal glands.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Obesity QRFP signaling modulates appetite; dysregulation may contribute to hyperphagia and obesity. Animal models and association studies (PMID: 16988026)
Type 2 Diabetes QRFP influences insulin secretion and glucose homeostasis. Expression studies in pancreatic islets (PMID: 17374880)
Metabolic Syndrome Altered QRFP levels linked to energy imbalance and metabolic dysfunction. Clinical association studies (PMID: 19401456)

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1Val) Missense <0.01% Likely loss of start codon; predicted loss of function
c.214C>T (p.Arg72Cys) Missense <0.01% Unknown; rare variant in population databases
Mutation functional classification

Loss of Function (LOF)

p.Met1Val likely abolishes translation initiation, leading to loss of QRFP peptide production.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations reported.

Gene Ontology (GO)

Protein Summary

The QRFP protein is a precursor that is cleaved into two bioactive peptides: QRFP-43 (43 amino acids) and QRFP-26 (26 amino acids). These peptides bind to the GPR103 receptor (QRFPR) to regulate appetite and energy expenditure. The protein is secreted and acts as a neuroendocrine modulator.
