Description

The PYY gene encodes Peptide YY, a 36-amino acid hormone secreted by L-cells in the distal gastrointestinal tract. It regulates appetite, gastric emptying, and pancreatic secretion. PYY acts via neuropeptide Y receptors (Y1, Y2, Y4, Y5) to reduce food intake and modulate energy homeostasis. Dysregulation is implicated in obesity, eating disorders, and gastrointestinal motility disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Obesity Reduced PYY levels lead to decreased satiety signaling and increased food intake Clinical studies show lower fasting and postprandial PYY in obese individuals (PMID: 12499395)
Anorexia Nervosa Elevated PYY contributes to excessive satiety and reduced caloric intake Plasma PYY levels are significantly higher in anorexia patients (PMID: 16390859)
Irritable Bowel Syndrome (IBS) Altered PYY secretion affects colonic motility and sensation Reduced PYY expression in colonic mucosa of IBS patients (PMID: 19834466)

Protein Summary

Peptide YY (PYY) is a 36-amino acid hormone processed to PYY(3-36), the major circulating form. It binds preferentially to Y2 receptors, inhibiting gastric motility and reducing appetite. PYY is secreted postprandially in proportion to caloric intake. Its anorexigenic effects make it a target for anti-obesity therapies.
