Description

PTK6 (Protein Tyrosine Kinase 6), also known as BRK (Breast Tumor Kinase), is a non-receptor tyrosine kinase belonging to the Src-related kinase family. It is predominantly expressed in epithelial tissues and plays a role in cell proliferation, differentiation, migration, and survival. PTK6 is frequently overexpressed or aberrantly activated in breast, ovarian, prostate, and colon cancers, where it can promote oncogenic signaling through pathways such as MAPK/ERK and PI3K/AKT. Its function is context-dependent, acting as both a tumor promoter and, in some settings, a tumor suppressor.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Overexpression and constitutive activation of PTK6 enhance proliferation and invasion via STAT3 and AKT signaling. PMID: 15626734; COSMIC; ClinVar
Ovarian cancer PTK6 overexpression correlates with poor prognosis and promotes cell migration through FAK phosphorylation. PMID: 18757403; COSMIC
Prostate cancer PTK6 is upregulated in prostate tumors and supports androgen receptor signaling and cell survival. PMID: 16951147; COSMIC
Colorectal cancer PTK6 expression is elevated in colon cancer and contributes to β-catenin/TCF transcriptional activity. PMID: 20010872; COSMIC

Mutation functional classification

Loss of Function (LOF)

Frameshift and start-loss mutations (e.g., p.Thr484Asnfs*2, p.Met1Val) are predicted to result in truncated or absent protein, reducing kinase activity.

Gain of Function (GOF)

No recurrent gain-of-function mutations have been reported in PTK6; overexpression and amplification are the primary oncogenic mechanisms.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for PTK6.

Pathways

MAPK/ERK signaling pathway
PI3K/AKT signaling pathway
STAT3 signaling pathway
FAK signaling pathway
Androgen receptor signaling pathway

Protein Summary

PTK6 (BRK) is a 451-amino-acid non-receptor tyrosine kinase with SH3, SH2, and catalytic domains. It is structurally similar to Src but lacks an N-terminal myristoylation signal, leading to distinct subcellular localization. PTK6 is activated by autophosphorylation at Tyr342 and can phosphorylate substrates such as STAT3, paxillin, and β-catenin. Its expression is largely restricted to epithelial cells, and dysregulation contributes to cancer progression by enhancing proliferation, migration, and survival signals.
