Description

PRKD3 (protein kinase D3) is a member of the protein kinase D (PKD) family of serine/threonine kinases. It is involved in signal transduction pathways regulating cell proliferation, differentiation, migration, and vesicle trafficking. PRKD3 is activated by diacylglycerol (DAG) and protein kinase C (PKC) and plays a role in cancer progression, immune response, and cardiac function.

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., p.Leu190Profs*5) lead to truncated, non-functional kinase domain.

Gain of Function (GOF)

Missense mutations such as p.Asn297Ser may enhance kinase activity, promoting oncogenic signaling.

Dominant Negative (DN)

No confirmed dominant-negative mutations reported in PRKD3.

Pathways

MAPK signaling pathway (Reactome: R-HSA-5683057)
VEGF signaling pathway (KEGG: hsa04370)
DAG/PKC signaling (Reactome: R-HSA-114608)
Regulation of actin cytoskeleton (KEGG: hsa04810)

Protein Summary

PRKD3 encodes a 912-amino acid serine/threonine kinase with a catalytic domain, two cysteine-rich zinc finger domains (C1a and C1b), and a pleckstrin homology (PH) domain. It is activated by DAG and PKC, localizes to the Golgi and plasma membrane, and regulates cell survival, motility, and secretion. Overexpression in cancers correlates with poor prognosis.
