Description

PRDX4 encodes peroxiredoxin 4, a member of the peroxiredoxin family of antioxidant enzymes. It reduces hydrogen peroxide and organic hydroperoxides, and is localized to the endoplasmic reticulum (ER) where it plays a key role in redox regulation, protein folding, and protection against oxidative stress. PRDX4 is involved in ER stress response, cell signaling, and apoptosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (breast, lung, prostate) Overexpression of PRDX4 promotes tumor growth by reducing oxidative stress and inhibiting apoptosis; also linked to chemoresistance. COSMIC, ClinVar, PubMed
Type 2 diabetes PRDX4 deficiency in pancreatic beta cells leads to increased oxidative stress and impaired insulin secretion. OMIM, PubMed
Neurodegenerative disorders (Alzheimer's, Parkinson's) Dysregulation of PRDX4 contributes to oxidative damage and protein misfolding in neurons. PubMed, ClinVar
Atherosclerosis PRDX4 modulates endothelial function and vascular inflammation via redox signaling. PubMed

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.586C>T (p.Arg196Cys) Missense <0.01% Reduced peroxidase activity; associated with increased oxidative stress
c.1A>G (p.Met1Val) Start loss <0.001% Loss of protein expression; likely pathogenic
c.364G>A (p.Gly122Ser) Missense 0.02% Altered substrate specificity; uncertain significance
Mutation functional classification

Loss of Function (LOF)

Mutations such as p.Met1Val lead to complete loss of protein expression, impairing antioxidant defense.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in PRDX4.

Dominant Negative (DN)

p.Arg196Cys may act in a dominant-negative manner by forming mixed disulfides with wild-type PRDX4, reducing overall activity.

Pathways

hsa04141 – Protein processing in endoplasmic reticulum
hsa04064 – NF-kappa B signaling pathway
hsa04211 – Longevity regulating pathway
hsa04931 – Insulin resistance

Protein Summary

Peroxiredoxin 4 (PRDX4) is a 271-amino acid antioxidant enzyme localized to the endoplasmic reticulum. It reduces H2O2 and organic hydroperoxides using thioredoxin as an electron donor. PRDX4 forms homodimers and contains a conserved Cys-Cys active site. It is critical for ER redox homeostasis, protein folding, and protection against oxidative damage. Dysregulation of PRDX4 is implicated in cancer, diabetes, and neurodegenerative diseases.
