Description
PLCB1 encodes phospholipase C beta 1, an enzyme that hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messengers inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG). This gene is involved in G protein-coupled receptor signaling and plays critical roles in neuronal development, synaptic plasticity, and cell proliferation. Mutations in PLCB1 are associated with infantile epileptic encephalopathy and have been implicated in various cancers.

Mutations & Variants
Hotspot Mutations
| Variant | Type | Frequency | Functional Description |
|---|---|---|---|
| c.1786C>T (p.Arg596*) | Nonsense | Rare | Loss of function; truncation of catalytic domain |
| c.2032G>A (p.Gly678Arg) | Missense | Rare | Likely loss of function; disrupts active site |

Mutation functional classification
Loss of Function (LOF)
Nonsense and missense mutations in the catalytic domain reduce or abolish PIP2 hydrolysis, leading to impaired neuronal signaling and epileptic encephalopathy.
Gain of Function (GOF)
Not well-documented; some cancer-associated variants may increase activity, but evidence is limited.
Dominant Negative (DN)
Not reported for PLCB1.

Protein Summary
Phospholipase C beta 1 (PLC-beta-1) is a 1216-amino acid protein that catalyzes the hydrolysis of PIP2 into IP3 and DAG. It contains a pleckstrin homology (PH) domain, EF-hand motifs, catalytic X and Y domains, and a C-terminal C2 domain. PLCB1 is activated by G alpha q subunits and is essential for calcium mobilization and protein kinase C activation. The protein is predominantly expressed in the brain and plays a key role in neuronal excitability and synaptic plasticity.
