Description
PLA2G5 encodes group V secretory phospholipase A2 (sPLA2-V), an enzyme that catalyzes the hydrolysis of membrane phospholipids to release arachidonic acid and lysophospholipids. This protein plays a key role in eicosanoid biosynthesis, inflammation, and lipid metabolism. It is expressed in various tissues and is implicated in atherosclerosis, asthma, and cancer progression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Atherosclerosis | sPLA2-V promotes LDL modification and foam cell formation via hydrolysis of phospholipids in arterial walls. | NCBI Gene, OMIM |
| Asthma | Increased sPLA2-V activity in airway epithelial cells contributes to eicosanoid-mediated inflammation and bronchoconstriction. | NCBI Gene, OMIM |
| Colorectal Cancer | Overexpression of PLA2G5 in tumor cells enhances prostaglandin E2 production, promoting cell proliferation and invasion. | COSMIC, NCBI Gene |

Protein Summary
The PLA2G5 protein (group V secretory phospholipase A2) is a 138-amino acid enzyme that hydrolyzes phospholipids at the sn-2 position, releasing arachidonic acid and lysophospholipids. It is secreted by inflammatory cells and acts extracellularly to modulate lipid signaling. The protein contains a conserved Ca2+-binding loop and a catalytic His-Asp dyad. Its expression is upregulated in inflammatory conditions and certain cancers.
