Description

PLA2G1B encodes a secreted phospholipase A2 enzyme primarily expressed in the pancreas. It catalyzes the hydrolysis of sn-2 fatty acyl bonds of phospholipids to release free fatty acids and lysophospholipids, playing a key role in dietary lipid digestion. The enzyme also participates in inflammatory signaling and has been implicated in pancreatic cancer and acute pancreatitis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Acute Pancreatitis Excessive activation of PLA2G1B within pancreatic acinar cells leads to autodigestion and inflammation PMID: 25921289
Pancreatic Cancer Overexpression of PLA2G1B promotes tumor cell proliferation and invasion via arachidonic acid metabolism PMID: 29371790
Crohn's Disease Altered PLA2G1B expression may contribute to intestinal inflammation through dysregulated eicosanoid production PMID: 21383956

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.331G>A (p.Gly111Arg) Missense <0.01% Reduced enzymatic activity; associated with familial pancreatitis risk
c.422C>T (p.Thr141Met) Missense <0.01% Altered substrate specificity; reported in pancreatic cancer
c.1A>G (p.Met1Val) Start loss <0.01% Loss of protein expression; rare variant
Mutation functional classification

Loss of Function (LOF)

Missense mutations (e.g., p.Gly111Arg) reduce catalytic activity, impairing lipid digestion and potentially increasing pancreatitis susceptibility.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in PLA2G1B.

Dominant Negative (DN)

No dominant-negative mutations described for PLA2G1B.

Pathways

Phospholipid metabolism (Reactome: R-HSA-1483206)
Arachidonic acid metabolism (KEGG: hsa00590)
Pancreatic secretion (KEGG: hsa04972)

Protein Summary

PLA2G1B is a 144-amino acid secreted protein (mature form: 125 residues) with a molecular weight of approximately 14 kDa. It contains a conserved Ca2+-binding loop and a catalytic His-Asp dyad. The enzyme is synthesized as a zymogen and activated by tryptic cleavage. It preferentially hydrolyzes phosphatidylcholine and phosphatidylethanolamine at the sn-2 position, releasing arachidonic acid and lysophospholipids. The protein is a member of the secreted phospholipase A2 family (sPLA2) and is abundant in pancreatic juice.
