Description

PKN3 (Protein Kinase N3) is a member of the protein kinase C (PKC) superfamily and encodes a serine/threonine kinase involved in cell proliferation, migration, and cytoskeletal organization. It is activated by Rho family small GTPases and plays a role in cancer progression, particularly in prostate and breast cancers. PKN3 is also implicated in insulin signaling and vascular development.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate Cancer PKN3 overexpression promotes tumor cell invasion and metastasis via Rho/ROCK signaling PMID: 21715324
Breast Cancer PKN3 knockdown reduces cell migration and invasion in triple-negative breast cancer cell lines PMID: 25605247
Insulin Resistance PKN3 modulates insulin-stimulated glucose uptake in adipocytes PMID: 19074966

Mutation functional classification

Loss of Function (LOF)

In-frame deletions in the kinase domain (e.g., p.Leu341del) are predicted to impair catalytic activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in PKN3.

Dominant Negative (DN)

No dominant-negative variants documented for PKN3.

Pathways

• Rho GTPase cycle (R-HSA-194840)
• Signaling by Rho family GTPases (R-HSA-194315)
• Regulation of actin cytoskeleton (KEGG:04810)
• PI3K-Akt signaling pathway (KEGG:04151)

Protein Summary

PKN3 is a 889-amino-acid serine/threonine kinase with an N-terminal regulatory region containing a Rho-binding domain and a C-terminal catalytic kinase domain. It is activated by binding to RhoA, RhoB, or RhoC, leading to autophosphorylation and subsequent phosphorylation of downstream targets such as vimentin and myosin light chain. PKN3 localizes to the cytoplasm and cell membrane and is involved in cytoskeletal remodeling, cell motility, and tumor invasion.
