Description

The PGP gene encodes phosphoglycolate phosphatase, an enzyme that catalyzes the dephosphorylation of 2-phosphoglycolate to glycolate and inorganic phosphate. This reaction is part of the 2-phosphoglycolate salvage pathway, which detoxifies 2-phosphoglycolate produced as a byproduct of DNA repair and other metabolic processes. The enzyme is also involved in the regulation of intracellular phosphate levels and has been implicated in cancer cell metabolism.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Altered PGP expression may affect cellular phosphate metabolism and promote tumor growth COSMIC; literature
2-Phosphoglycolate accumulation disorders Deficiency in PGP leads to accumulation of 2-phosphoglycolate, potentially causing cellular toxicity OMIM; literature

Protein Summary

Phosphoglycolate phosphatase (PGP) is a 32 kDa enzyme that belongs to the haloacid dehalogenase (HAD) superfamily. It specifically hydrolyzes 2-phosphoglycolate to glycolate and phosphate. The protein is localized in the cytoplasm and is expressed in multiple tissues, with highest levels in liver and kidney. PGP plays a critical role in detoxifying 2-phosphoglycolate, which can inhibit glycolysis and DNA repair if accumulated.
