Description
The PGC gene encodes progastricsin, also known as pepsinogen C, a precursor of the gastric aspartic protease gastricsin. This enzyme is secreted by gastric chief cells and is involved in protein digestion in the stomach. PGC expression is often downregulated in gastric cancer and other gastric disorders, making it a potential biomarker for gastric mucosal integrity and differentiation.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Gastric Cancer | Loss of PGC expression is associated with gastric carcinogenesis; reduced progastricsin levels reflect loss of chief cell differentiation. | Multiple studies; NCBI Gene, PubMed |
| Atrophic Gastritis | Decreased PGC expression correlates with gastric atrophy and intestinal metaplasia. | ClinVar, literature |
| Peptic Ulcer Disease | Altered pepsinogen levels (including PGC) are observed in patients with peptic ulcers. | OMIM, literature |

Protein Summary
Progastricsin (PGC) is a 388-amino-acid precursor protein that is proteolytically cleaved to yield active gastricsin, an aspartic protease. It is primarily expressed in gastric chief cells and secreted into the stomach lumen. The enzyme functions optimally at acidic pH and contributes to the digestion of dietary proteins. Reduced expression is a hallmark of gastric atrophy and gastric cancer.
