Description

PGA3 (Pepsinogen A3) is a protein-coding gene located on chromosome 11q12.2. It encodes a precursor of pepsin A, an aspartic protease secreted by gastric chief cells. Pepsinogen A3 is activated to pepsin A in the acidic gastric environment and plays a key role in protein digestion. The gene is part of the pepsinogen A gene cluster and is frequently used as a biomarker for gastric mucosal status and gastric cancer risk.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Gastric Cancer Altered expression of PGA3 is associated with gastric carcinogenesis; reduced pepsinogen levels are used as a serological marker for atrophic gastritis and gastric cancer risk. ClinVar, COSMIC
Atrophic Gastritis Decreased PGA3 expression reflects loss of gastric chief cells in chronic atrophic gastritis. ClinVar, NCBI
Peptic Ulcer Disease Pepsinogen levels, including PGA3, are altered in peptic ulcer disease, though direct causal mechanisms are not fully established. NCBI

Protein Summary

Pepsinogen A3 is a 388-amino-acid precursor protein (UniProt P00790) that is cleaved in the acidic gastric lumen to form active pepsin A. The active enzyme is an aspartic protease with broad specificity for hydrophobic residues. It is primarily expressed in gastric chief cells and secreted into the stomach lumen. The protein structure includes a signal peptide (residues 1-16) and an activation peptide (residues 17-58) that is removed during activation.
