Description

PEDS1 encodes plasmalogen synthase 1, an enzyme that catalyzes the final step in plasmalogen biosynthesis, converting 1-alkyl-2-acyl-sn-glycerol to plasmenylethanolamine. Plasmalogens are ether phospholipids critical for membrane integrity, signaling, and antioxidant defense. The gene is located on chromosome 11q13.4 and is expressed in multiple tissues, with highest levels in brain, heart, and skeletal muscle.

Mutation functional classification

Loss of Function (LOF)

Mutations leading to truncated or inactive enzyme reduce plasmalogen levels, causing peroxisomal dysfunction.

Gain of Function (GOF)

Not reported; no activating mutations known.

Dominant Negative (DN)

Not reported; no dominant-negative mechanisms described.

Pathways

• Ether lipid metabolism (Reactome: R-HSA-1483206)
• Plasmalogen biosynthesis (KEGG: map00564)

Protein Summary

Plasmalogen synthase 1 is a 595-amino acid transmembrane protein localized to the endoplasmic reticulum. It catalyzes the formation of plasmenylethanolamine from 1-alkyl-2-acyl-sn-glycerol and CDP-ethanolamine. The protein contains a conserved domain for glycerophosphodiester phosphodiesterase activity. Defects in this enzyme lead to reduced plasmalogen levels, which are associated with peroxisomal disorders and altered lipid metabolism in cancer.
