Description
PEBP1 encodes phosphatidylethanolamine binding protein 1, also known as Raf kinase inhibitory protein (RKIP). It functions as a negative regulator of the MAPK/ERK signaling cascade by binding to Raf-1 and inhibiting its activation. Additionally, PEBP1 modulates G protein-coupled receptor signaling and has roles in spermatogenesis, neuroprotection, and metastasis suppression. The protein is widely expressed, with highest levels in brain, testis, and prostate.

Protein Summary
PEBP1 is a 187-amino acid protein (21 kDa) that adopts a beta-sandwich fold with a conserved ligand-binding pocket. It binds phosphatidylethanolamine and inhibits Raf-1 kinase activity, thereby suppressing the MAPK/ERK pathway. The protein also functions as a precursor of hippocampal cholinergic neurostimulating peptide (HCNP) and is involved in acetylcholine synthesis. Post-translational modifications include phosphorylation at Ser96, which modulates its inhibitory activity.
