Description

PDGFD encodes platelet-derived growth factor D, a member of the PDGF/VEGF family. The protein is synthesized as a latent precursor that requires proteolytic cleavage to become active. It binds to PDGFRB and plays roles in angiogenesis, wound healing, fibrosis, and tumor growth. Overexpression is linked to various cancers and fibrotic diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Glioma PDGFD promotes tumor angiogenesis and proliferation via PDGFRB signaling. PubMed: 19632184
Idiopathic Pulmonary Fibrosis PDGFD is upregulated in lung fibroblasts, driving fibrosis through PDGFRB activation. PubMed: 21525062
Ovarian Cancer PDGFD overexpression correlates with poor prognosis and increased angiogenesis. PubMed: 22952446

Pathways

PDGF signaling pathway (Reactome: R-HSA-186797)
Signaling by Receptor Tyrosine Kinases (Reactome: R-HSA-9006934)
Focal Adhesion-PI3K-Akt-mTOR pathway (KEGG: hsa04510)

Protein Summary

PDGFD is a 370-amino-acid secreted growth factor containing a CUB domain and a PDGF/VEGF homology domain. Proteolytic removal of the CUB domain releases the active growth factor, which binds to PDGFRB, activating downstream signaling cascades (MAPK, PI3K/Akt). It is implicated in developmental processes, tissue repair, and pathological conditions including fibrosis and tumor angiogenesis.
