Description
PCLAF (PCNA clamp associated factor) encodes a protein that binds to proliferating cell nuclear antigen (PCNA) and regulates DNA replication and repair. It is involved in cell cycle progression, DNA damage response, and chromatin remodeling. Overexpression is observed in multiple cancers and correlates with poor prognosis.

Protein Summary
PCLAF is a 15 kDa nuclear protein that directly interacts with PCNA via a conserved PIP-box motif. It modulates PCNA loading onto DNA, influences replication fork progression, and participates in DNA damage bypass. Its expression is cell-cycle regulated, peaking in S phase. In cancer, PCLAF overexpression drives proliferation and therapy resistance.
