Description

PCDH1 (protocadherin 1) is a member of the cadherin superfamily of calcium-dependent cell adhesion molecules. It encodes a non-clustered protocadherin with 7 extracellular cadherin repeats, a transmembrane domain, and a cytoplasmic tail. PCDH1 is expressed in multiple tissues including lung, brain, and kidney, and plays a role in cell-cell adhesion, neuronal development, and airway epithelial integrity. Genetic variants in PCDH1 have been associated with asthma and bronchial hyperresponsiveness.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Asthma PCDH1 variants may alter airway epithelial cell adhesion and barrier function, contributing to bronchial hyperresponsiveness Association studies (Koppelman et al. 2009, NCBI)
Bronchial Hyperresponsiveness Loss of PCDH1 function in airway epithelium may increase sensitivity to environmental triggers Genetic linkage and association (OMIM #603626)
Lung Function Decline PCDH1 expression changes in lung tissue correlate with reduced FEV1 GWAS and expression studies (NCBI Gene)

Loss of Function (LOF)

Missense variants in extracellular cadherin repeats may disrupt calcium binding and adhesion (e.g., p.Gly420Arg).

Gain of Function (GOF)

Not reported for PCDH1.

Dominant Negative (DN)

Not established; PCDH1 likely functions as a homodimer, but dominant-negative effects have not been documented.

Protein Summary

Protocadherin 1 is a 1067-amino acid transmembrane protein belonging to the cadherin superfamily. It contains 7 extracellular cadherin repeats, a single transmembrane domain, and a cytoplasmic region with conserved motifs. The protein mediates homophilic cell-cell adhesion in a calcium-dependent manner. PCDH1 is expressed in airway epithelium, where it contributes to barrier integrity, and in neural tissues where it may influence synaptic development. Alternative splicing generates multiple isoforms.
