Description

PANK3 encodes pantothenate kinase 3, an enzyme that catalyzes the first and rate-limiting step in coenzyme A (CoA) biosynthesis: the phosphorylation of pantothenate (vitamin B5) to 4'-phosphopantothenate. This gene is ubiquitously expressed and plays a critical role in cellular metabolism, fatty acid oxidation, and energy production. Mutations in PANK3 have been associated with metabolic disorders and neurodegeneration.

Protein Summary

Pantothenate kinase 3 is a 370-amino acid protein belonging to the pantothenate kinase family. It is localized in the cytoplasm and cytosol, where it phosphorylates pantothenate using ATP. The enzyme is essential for maintaining cellular CoA levels, which are critical for fatty acid metabolism, the tricarboxylic acid cycle, and acetylation reactions. PANK3 is widely expressed, with highest levels in liver and brain.
