Description

PABPC4 encodes a member of the poly(A)-binding protein family, which binds to the poly(A) tail of mRNA and plays a critical role in mRNA stability, translation initiation, and polyadenylation. The protein is predominantly cytoplasmic and is involved in regulating gene expression at the post-transcriptional level. PABPC4 is also implicated in cellular responses to viral infections and in cancer progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Overexpression of PABPC4 enhances mRNA stability of oncogenes, promoting cell proliferation. PMID: 25691885
Colorectal cancer PABPC4 upregulation correlates with poor prognosis and increased metastatic potential. PMID: 29736038
HIV-1 infection PABPC4 interacts with HIV-1 Rev protein to facilitate nuclear export of viral mRNA. PMID: 22114356
Influenza A infection PABPC4 is hijacked by influenza virus to enhance viral mRNA translation. PMID: 23345592

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations in PABPC4 are predicted to cause loss of function by truncating the protein, impairing mRNA binding and translation regulation.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in PABPC4.

Dominant Negative (DN)

No evidence for dominant-negative effects in PABPC4.

Pathways

• mRNA surveillance pathway (KEGG: hsa03015)
• RNA transport (KEGG: hsa03013)
• Translation factors (Reactome: R-HSA-156842)

Protein Summary

PABPC4 is a 644-amino acid protein containing four RNA recognition motifs (RRMs) and a C-terminal domain. It binds with high affinity to poly(A) tails of mRNA, promoting circularization of mRNA via interaction with eIF4G and enhancing translation initiation. PABPC4 also shuttles between nucleus and cytoplasm and is involved in stress granule assembly. Its expression is ubiquitous but enriched in testis and lymphoid tissues.
