Description

P2RY8 encodes a G protein-coupled receptor (GPCR) for extracellular nucleotides, primarily ATP and ADP. It is expressed on germinal center B cells and mediates chemotaxis and cell positioning via the CXCL13-CXCR5 axis. P2RY8 signaling is critical for B cell migration within lymphoid follicles and is implicated in diffuse large B-cell lymphoma (DLBCL) and other B-cell malignancies.

Loss of Function (LOF)

Loss-of-function mutations (e.g., start codon loss) impair receptor expression and B cell migration.

Gain of Function (GOF)

P2RY8-CXCR4 fusions result in gain-of-function via constitutive CXCR4 signaling.

Dominant Negative (DN)

No dominant-negative mutations reported.

Protein Summary

P2RY8 is a 377-amino acid GPCR with seven transmembrane domains. It binds extracellular nucleotides and couples to Gq/11 proteins to activate phospholipase C and calcium mobilization. In germinal center B cells, P2RY8 promotes chemotaxis toward CXCL13 and helps position B cells within the light zone. The receptor is also expressed in T cells and dendritic cells, where it modulates immune responses.
